Endomembrane System and Endoplasmic Reticulum Functions

Inside eukaryotic cells, many membrane-bound parts work as one connected system called the endomembrane system. Its most important member is the endoplasmic reticulum (ER), a network of folded membranes.

This topic matters because ER makes proteins and lipids, detoxifies substances and starts the pathway that sends materials to Golgi, lysosomes and cell surface.

Key Concepts

Endomembrane system includes nuclear envelope, ER, Golgi apparatus, lysosomes, vacuoles and plasma membrane (functionally linked). ER is of two types: rough ER (RER) with ribosomes, making proteins; smooth ER (SER) without ribosomes, making lipids and detoxifying drugs. ER lumen is internal space where folding occurs. Transport vesicles carry products from ER to Golgi.

How It Works

  1. Ribosomes on RER synthesise proteins that enter ER lumen.
  2. Proteins fold and get modified inside ER; wrongly folded proteins are removed.
  3. SER synthesises lipids, steroid hormones and breaks down toxins in liver cells.
  4. Vesicles bud from ER and fuse with Golgi for further processing and sorting.
  5. Final products go to lysosomes, membrane or secretion outside cell.

Worked Example

A pancreatic cell makes digestive enzyme protein. Ribosome on RER makes a chain of 300 amino acids. It enters ER, folds and gets packaged in a vesicle. If cell produces about 1,000 vesicles per hour each carrying many enzyme molecules, total secretion becomes very large. Step by step, ER starts production, Golgi finishes and vesicles release enzymes into pancreatic duct — showing division of labour.

Common Misconceptions

  • ER and Golgi are same organelle. ER makes and initially processes; Golgi modifies, sorts and dispatches.
  • Smooth ER has ribosomes. Smooth ER lacks ribosomes; rough ER has them.
  • All proteins are made on ER. Many cytoplasmic proteins are made on free ribosomes, not ER.

Real-World Applications

Liver SER detoxifies alcohol and medicines; excess use can damage it. Insulin and antibody production depend on RER-Golgi pathway. Genetic diseases of protein folding (like cystic fibrosis partly) involve ER quality control. Biotechnology uses cell ER pathways to produce therapeutic proteins.

The endomembrane system is the cell's factory and postal network, with ER as the main production site for proteins and lipids.
