Named Reactions: Wittig, Diels-Alder, Claisen & Perkin

Organic chemistry honours certain transformations with names, and five of them appear constantly in exams: the Wittig reaction builds alkenes, the Diels–Alder reaction builds six-membered rings, the Claisen condensation builds β-keto esters, the Dieckmann condensation closes rings the same way, and the Perkin reaction builds cinnamic acid. Each name is shorthand for a reliable bond-forming pattern.

These reactions matter because they are the standard tools for constructing carbon skeletons — alkenes from carbonyls, rings from dienes, and extended conjugation from aldehydes. Examiners test them by asking for the product, the reagents, or the mechanism's key step.

Key Concepts

  • Wittig reaction: a phosphorus ylide (Ph₃P⁺−C⁻HR) reacts with an aldehyde or ketone to give an alkene + triphenylphosphine oxide (Ph₃P=O).
  • Diels–Alder reaction: a conjugated diene + a dienophile (alkene) undergo concerted [4+2] cycloaddition to give a substituted cyclohexene; stereospecific.
  • Claisen condensation: two ester molecules + strong base (NaOEt) → a β-keto ester + alcohol; e.g. ethyl acetate → ethyl acetoacetate.
  • Dieckmann condensation: the intramolecular Claisen of a diester → a cyclic β-keto ester (5- or 6-membered rings form best).
  • Perkin reaction: an aromatic aldehyde + an acid anhydride + the sodium salt of the acid → an α,β-unsaturated acid; e.g. benzaldehyde → cinnamic acid.

The Wittig ylide (from alkyl halide + Ph₃P + strong base) attacks the carbonyl carbon, and the oxaphosphetane intermediate collapses to the alkene. Diels–Alder needs the diene in s-cis conformation and is accelerated by electron-withdrawing groups on the dienophile. Claisen needs an ester with α-hydrogens and a matching alkoxide base; Perkin proceeds via aldol-type condensation and dehydration.

Formulas

  • Wittig: Ph₃P=CHR + R′₂C=O → R′₂C=CHR + Ph₃P=O. Converts C=O into C=C at the same carbon.
  • Diels–Alder: diene (4π) + dienophile (2π) → cyclohexene derivative. Follows the endo rule: substituents on the dienophile end up cis to the new double bond's underside.
  • Claisen: 2CH₃COOC₂H₅ → CH₃COCH₂COOC₂H₅ + C₂H₅OH (with NaOEt). Product: ethyl acetoacetate.
  • Perkin: C₆H₅CHO + (CH₃CO)₂O → C₆H₅CH=CHCOOH (with CH₃COONa). Benzaldehyde (106) gives cinnamic acid (148 g mol⁻¹).

Worked Example

The Perkin reaction is run with 10.6 g of benzaldehyde and excess acetic anhydride. What is the theoretical yield of cinnamic acid? Step 1: moles of benzaldehyde = 10.6 ÷ 106 = 0.10 mol. Step 2: the equation is 1:1, so 0.10 mol of cinnamic acid forms. Step 3: molar mass of cinnamic acid, C₆H₅CH=CHCOOH = 148 g mol⁻¹. Step 4: theoretical mass = 0.10 × 148 = 14.8 g. So the maximum obtainable cinnamic acid is 14.8 g; the percentage yield compares the actual isolated mass to this figure.

Common Misconceptions

  • "The Wittig reagent is just a Grignard reagent." No — the ylide has a positively charged phosphorus stabilising the adjacent carbanion; Grignard reagents add to carbonyls to give alcohols, while Wittig gives alkenes.
  • "Diels–Alder works with any diene shape." Wrong — the diene must adopt the s-cis conformation; locked s-trans dienes (like (E,E)-hexadiene frozen open) cannot react.
  • "Claisen condensation works on esters without α-hydrogens." It does not — the mechanism needs α-proton abstraction to form the enolate nucleophile.

Real-World Applications

  • Wittig: industrial synthesis of vitamin A and carotenoids uses Wittig steps to build their long alkene chains.
  • Diels–Alder: a key ring-forming step in the synthesis of steroids, insecticides and many natural products.
  • Claisen/Dieckmann: β-keto esters like ethyl acetoacetate are versatile building blocks for heterocycles and drugs; Perkin's cinnamic acid derivatives appear in fragrances and UV-absorbing sunscreens.

Wittig turns carbonyls into alkenes, Diels–Alder snaps dienes and dienophiles into rings, Claisen/Dieckmann forge β-keto esters, and Perkin extends aldehydes into cinnamic acids. Learn one reagent set and one product per reaction.
