Why do 90% of drug candidates fail? Where? Step 1: Most fail in Phase II/III — efficacy or safety in humans. Step 2: Animal models don't perfectly predict humans. Step 3: Toxicity (liver, heart) or no better than existing drugs. Step 4: Pipeline is a funnel — thousands in, one drug out!
